| Abstract | Provided are methods and compositions for generating or maintaining human iPS cells in culture including a use of a low osmolality medium comprising a base medium and supplements comprising a LIF polypeptide, a GSK3 inhibitor, and a MEK inhibitor; wherein the base medium has an osmolality of about 180 mOsm/kg to about 250 mOsm/kg. In an embodiment, the human iPS cells cultured in the low osmolality medium express phenotypes, gene expression profiles, or markers characteristic to a naive state. |
