| Abstract | Provided is a gene mutation introduction method with improved sequence-specificity comprising a step of expressing a fusion protein comprising a sequence-specific DNA cleavage enzyme and a nuclear receptor under control of an expression-inducing promoter, a step of forming a double-strand DNA break in genomic DNA in a sequence-specific manner by transferring the fusion protein into nucleus, and a step of repairing the double-strand DNA break by utilizing a DNA break repair mechanism and thereby introducing a gene mutation into the vicinity of the double-strand DNA break. |
