Abstract

Provided is a method for selectively inducing a sub-population of definitive endoderm cells selected from the group consisting of anterior-domain-of-anterior-definitive-endoderm (AADE) cells, posterior-domain-of-anterior-definitive-endoderm (PADE) cells, and posterior-definitive-endoderm (PDE) cells, from pluripotent stem cells, comprising steps of; (1) culturing the pluripotent stem cells in a medium containing Activin A and a GSK3β inhibitor, to obtain early anterior primitive streak cells or late anterior primitive streak cells, and (2) culturing the early anterior primitive streak cells or late anterior primitive streak cells obtained in step (1) in a medium containing Activin A, and either or not containing a BMP inhibitor, to obtain AADE cells, PADE cells, or PDE cells. Also provided is a method for selectively inducing primitive gut cells selected from the group consisting of anterior foregut (AFG) cells, posterior foregut (PFG)cells, and midhindgut (MHG) cells by culturing the sub-population of definitive endoderm cells obtained in step (2) of the aforementioned method in a medium not-containing differentiation factors.
