Abstract

The present invention provides a method for producing human primordial germ cell (PGC)-like cells from human pluripotent stem cells with high efficiency and reproducibility, comprising (i) a step of culturing human pluripotent stem cells in a medium containing Activin A and a GSK3β inhibitor to induce mesoderm-like cells, and (ii) a step of culturing the mesoderm-like cells obtained in step (i) in a medium containing BMP. The invention further relates to a method for isolating human PGC-like cells, comprising (iii) a step of selecting the cells positive for at least one cell surface maker selected from the group consisting of PECAM (CD31), INTEGRINα6 (CD49f), INTEGRINβ3 (CD61), KIT (CD117), EpCAM, PODOPLANIN and TRA1-81, from the cells obtained in the step (ii).
