Abstract

This invention provides a method for producing an airway epithelial cell from a pluripotent stem cell comprising steps of; (1) culturing a pluripotent stem cell in a medium containing Activin A and a GSK3β inhibitor, (2) culturing the cell obtained in (1) in a medium containing a BMP inhibitor and a TGFβ inhibitor, (3) culturing the cell obtained in (2) in a medium containing BMP4, retinoic acid, and a GSK3β inhibitor, (5) 3D culturing the ventral anterior foregut cell obtained after step (3) in a medium containing a GSK3β inhibitor, FGF10, and a ROCK inhibitor, and (6) 3D culturing the central airway epithelial progenitor cell obtained in (5) in a medium containing a ROCK inhibitor. The method may further comprise the step (4); culturing the ventral anterior foregut cell obtained after step (3) in a medium containing a GSK3β inhibitor and FGF10 to induce an airway epithelial progenitor cell.
