Abstract

Provided is a novel mRNA switch which enables the construction of a precise translation regulation circuit and can be used as a translation regulation module having versatility and orthogonality. In one embodiment, the mRNA switch of the invention comprises an artificial mRNA molecule which comprises (i) a nucleic acid sequence capable of being recognized specifically by an input protein comprising Cas protein or a variant thereof and (ii) a nucleic acid sequence encoding an output protein, wherein the nucleic acid sequence (i) is located on the 5'- or 3'-side of the nucleic acid sequence (ii) and the nucleic acid sequences (i) and (ii) are operably linked to each other. According to the invention, a mRNA switch having a translation control module that does not interfere with other modules can be obtained, enabling a wide variety of applications such as safer and efficient cell programming.
