Abstract | Provided is a mRNA comprising (i) a nucleic acid sequence specifically recognized by a miRNA or a protein, and (ii) a nucleic acid sequence of a coding region of a marker protein, wherein the mRNA contains N1-methyl-pseudouridine nucleotide. When introduced into a cell, the said mRNA functions as a mRNA switch, either on- or off-switch, to control the expressions of a marker protein in response to an intracellular miRNA or protein. The mRNA switch of the invention shows an elevated expression level when introduced into a cell, and hence in an embodiment, a large increase in the expression of a marker protein can be achieved relative to the prior art, especially for a membrane protein or a secretory protein. |
